a) What features do you want the AraPβ regulatory region to have from a commercial perspective? Justify your answer.

b) If you were working as a biomedical engineer for this company, which mutant would you recommend has the most commercial value? Justify.

c) Using the genetic information provided for the AraPβ1 and AraPβ4 reporter constructs (nucleotide sequences), describe how their observed phenotype can be attributed to a mutation in the respective genotype of these two reporter constructs. To determine this, you need to:

    • compare the phenotype in the mutants with the phenotype of the wildtype, for each reporter construct, to determine how the mutation presents, phenotypically.
    • check for mutations in the regulatory regions of the gene - this would include the AraC binding site and the promoter region. This is so that you can locate a genetic mutation that is responsible for a difference in the phenotype.
    • predict the impact of these genetic mutations on the transcription of the reporter gene and relate this to the phenotype observed in the lab.
