To determine this, you need to:
- compare the phenotype in the mutants with the phenotype of the wildtype, for each reporter construct, to determine how the mutation presents, phenotypically.
- check for mutations in the regulatory regions of the gene - this would include the AraC binding site and the promoter region. This is so that you can locate a genetic mutation that is responsible for a difference in the phenotype.
- predict the impact of these genetic mutations on the transcription of the reporter gene and relate this to the phenotype observed in the lab.
